Agarose particles are micro- to nanoscale beads or granules derived from agarose, a linear polysaccharide extracted from red algae. Owing to their hydrophilic nature, biocompatibility, and tunable porosity, agarose particles serve as versatile matrices in biochemical separation, immobilization techniques, and biomedical applications.

  • Structure and Composition:
    • Chemical Structure: Agarose is composed of repeating agarobiose units—a disaccharide of D-galactose and 3,6-anhydro-L-galactopyranose—linked via alternating α-1,3 and β-1,4 glycosidic bonds. Its linear, unbranched chain structure allows the formation of a three-dimensional network upon cooling from a heated solution.
  • Preparation and Surface Engineering:
    • Synthesis Methods: Common fabrication techniques include:
      • Emulsion Polymerization: An aqueous solution of agarose is emulsified in an oil phase to form droplets, which are then cooled to solidify into microspheres. Particle size is controlled by adjusting emulsification parameters (e.g., stirring speed, surfactant concentration).
      • Dripping or Atomization: Agarose solution is dispensed dropwise into a cold oil or saline bath, forming gelled beads upon contact.
  • Physical and Chemical Properties:

    • Biocompatibility: Inert and non-toxic, agarose does not interfere with biological molecules, preserving their structure and function—a key property for biomedical and biochemical applications.
    • Thermal Reversibility: Agarose gels are thermoreversible; they melt upon heating and re-gel upon cooling. This property is exploited in forming and processing particles under mild conditions.

    • Chromatography Media: Agarose particles form the basis of many gel filtration (size-exclusion) and affinity chromatography matrices (e.g., Sepharose). Their porous structure enables separation based on molecular size or specific binding interactions when functionalized with ligands.
